dc.description.abstract | A procedure has been developed for the isolation of the enterokinase inhibitors from the peanut seed. A peanut extract was subjected to affinity chromatography using an anhydrotrypsin Sepharose column, followed by ion exchange chromatography on a carboxymethyl-cellulose column in the presence of 8M urea. The latter step of the isolation scheme was developed only after obtaining a series of contradictory results from the pH 8.9 and pH 4.3 disc gels and isoelectric focusing gels, with and without urea. These gels suggested that upon aggregation the charge properties of the peanut inhibitors change greatly. The three peanut enterokinase inhibitors isolated were found to be positively charged by ion exchange chromatography in the presence of 8M urea, possessed a molecular weight of 8000 daltons and were single chain proteins. | |
