|Abstract:||The lyretail swordtail fish is a dominant mutant trait of common swordtail (Xiphophorus helleri). Three main approaches were used to identify the DNA markers to distinguish homozygous dominant lyretail from heterozygous lyretail. 1) Microsatellite primers, 2) Random amplified polymorphic DNA (RAPD), and 3) Suppression subtractive hybridization (SSH) cDNA library. One of the microsatellite primers, KonT38, revealed 250-bp DNA fragment in both common and heterozygous lyretails, but not in homozygous lyretails. Therefore this DNA fragment may be related to the for a recessive allele for lyretail phenotype. More than 650 reactions of RAPD showed many polymorphic DNA fragments. However, specific banding patterns related to the lyretail genotypes, which could be used as DNA markers, did not appear. A number of differentially expressed genes in the lyretail and common fins have also been identified through SSH analyses. The differentially expressed cDNAs isolated from the lyretail library include four genes for cell proliferation, while the cDNAs identified from common fin include genes for regulation of cell proliferation and inhibition of gene expressions.|
